Workflow for the direct identification of antigen-specific B-cells using a novel microfluidic platform. Using a unique cell microwell culture plate, B-cells can be seeded, cultured and analysed for their ability to recognise target antigens. After automated recovery of antigen-specific B cells, a single-cell PCR workflow amplifies variable regions of VH/VL antibodies. Amplicons can be sequenced, and the resulting data analysed through an in-house developed pipeline. The sequences can then be used to produce recombinant antibodies in vitro.
